Just ordered this speaker with my amazon gift card balance. What would be the best way to receive a refund for this item?
Can I use empty box or is the speaker too heavy?

It would be too heavy to EB, perhaps the most straightforward method would be DNA.

As mentioned, It's too heavy for the missing Item method and even with a weight substitute, too big to box without showing signs of tampering at the collection point.

You can opt for the DNA or wrong Item received.

Claimed it didn't arrive, they gave me an amazon giftcard for 5 euro for the inconvenience and starten an investigation with the courrier service, But due to corona they didn't ask my signature

No, I work for the same courrier. They sign it theirselves because of the COVID-19. And they gave it to my brother instead of me.
